In your cover letter, please explain why you decided to join this field, and what makes you uniquely qualified for this position? Try to be original, because it gets pretty boring reading the same responses over and over. "I just love software" is nice, but it really doesn't tell me anything. Dig a little deeper.
If you are applying for a developer position, please list the programming languages, technologies, and operating systems you are an expert in. If you've popped the top on Java, but never really done any heavy duty development with it, please don't list it. Even the best people I know are usually experts in 1 to 3 development tools. If you list 20, don't expect me to read any further.
One more thing. I do not hire trainees, junior developers, freshers, wannabe's, or any other term you might have heard to describe people lacking real-world experience. You must have a minimum of 7 years of real-world software development experience, preferably in connection with large, enterprise class applications.
